ABSTRACT

OBJECTIVE@#To investigate the role of acetylated modification induced by coactivator p300 in lipopolysaccharide (LPS)- induced inflammatory mediator synthesis and its molecular mechanism.@*METHODS@#Agilent SurePrint G3 Mouse Gene Expression V2 microarray chip and Western blotting were used to screen the molecules whose expression levels in mouse macrophages (RAW246.7) were correlated with the stimulation intensity of LPS. Electrophoretic mobility shift assay (EMSA) and chromatin immunoprecipitation (chip-qPCR) were used to verify the binding of the molecules to the promoters of IL-6 and TNF-α genes. The effects of transfection of RAW246.7 cells with overexpression or interfering plasmids on IL-6 and TNF-α synthesis were evaluated with ELISA, and the binding level of the target molecules and acetylation level of H3K27 in the promoter region of IL-6 and TNF-α genes were analyzed by chromatin immunoprecipitation sequencing technique (chip-seq).@*RESULTS@#Gene microarray chip data and Western blotting both confirmed a strong correlation of p300 expression with the stimulation intensity of LPS. Immunocoprecipitation confirmed the binding between p300 and c-myb. The results of EMSA demonstrated that c-myb (P < 0.05), but not p300, could directly bind to the promoter region of IL-6 and TNF-α genes; p300 could bind to the promoters only in the presence of c-myb (P < 0.05). The expressions of p65, p300 and c-myb did not show interactions. Both p300 overexpression and LPS stimulation could increase the level of promoter-binding p300 and H3K27 acetylation level, thus promoting p65 binding and inflammatory gene transcription; such effects were obviously suppressed by interference of c-myb expression (P < 0.05). Interference of p65 resulted in inhibition of p65 binding to the promoters and gene transcription (P < 0.05) without affecting p300 binding or H3K27 acetylation level.@*CONCLUSION@#LPS can stimulate the synthesis of p300, whose binding to the promoter region of inflammatory genes via c-myb facilitates the cohesion of p65 by inducing H3K27 acetylation, thus promoting the expression of the inflammatory genes.

ABSTRACT

OBJECTIVE@#To develop a universal screwdriver for sealing the end of the central hole of the femoral interlocking intramedullary nail, so as to shorten the operation time of the tail cap implantation of the intramedullary nail and improve the accuracy of implantation.@*METHODS@#Total 77 patients with intertrochanteric fractures underwent femoral interlocking intramedullary nail (FIIN) surgery from June 2018 to June 2019. There were 28 males and 49 females, aged 55 to 80 (76.22± 7.32) years old, and course of disease was 20 to 40 h. All patients were divided into universal screwdriver group (39 cases) and ordinary screwdriver group (38 cases) according to whether the self-developed universal screw was applicable during the operation. The blood loss during tail cap implantation, the time of tail cap implantation, the success rate of one-time implantation, and the postoperative curative effect were compared between two groups.@*RESULTS@#All patients were followed up for 12 to 36 months, with an average of(20.00±6.38) months. The bleeding volume and the time of tail cap implantation in the universal screwdriver group were significantly lower thanthose in the ordinary screwdriver group (@*CONCLUSION@#The universal screwdriver is easy to operate during the operation when using the cap of the femoral intramedullary nail, the operation time is shortened, the amount of bleeding is reduced, and the treatment effect is satisfactory.

ABSTRACT

The expression status of estrogen receptor (ER) and MRI features of breast cancer were introduced,which included needle-like edge,ring enhancement,types of time-to-intensity curve (TIC) as well as the relationship between ADC values.The breast cancer patients with expression statuses had different MRI features including MRI morphology,enhancement state,enhancement mode and diffusion,and the main cause was that ER positive expression suppressed neovascularization to influence the focal fibrosis,necrosis,growth rate,perfusion and etc.Hormone level was pointed out to be evaluated by morphological manifestations of MRI dynamic enhancement and ADC value of diffusion weighted imaging,so that valuable information could be provided for the prognosis evaluation,treatment planning as well as the studies on ER expression status and TIC types.

ABSTRACT

Objective To investigate the diagnostic efficiency and result of prostate biopsy for patients with t -PSA between 4.0 and 10 ng/ml.Methods This analysis was based on 20 qualified research papers from such lectronic databases as PubMed, MEDLINE, EMBASE and Cochrane from January 2010 until September 2017.Data extracted was analyzed using classic Meta-analysis with R software .The random or fixed effect model analysis was used to estimate the rate.Heterogeneity was analyzed using I 2 statistic.Results Totally 5481 patients were included in the 20 research papers. The positive rate of prostate biopsy was 20.6%, with t-PSA between 4.0 and 10 ng/ml, which was higher than the rate in the data from CUA Guide(2014).The difference was statistically significant .Conclusion Patients should be subjected to prostate biopsy if their t-PSA ranges 4.0 from 10 ng/ml regardless of the rate of f/t-PSA.The Gleason grade is relatively low when PSA is in the gray area , and the risk is also low.

ABSTRACT

Objective: To optimize the technology of freeze-dried powder of various fractions from lumbricus (Pheretima) by Box-Behnken design and response surface method. Methods: Using single factor experiment with Box-Behnken response surface method, freeze-dried rate as the index, the effects of sublimation temperature, sublimation time, analytical drying temperature, and drying time on the freeze-drying process were observed; The freeze-drying process of the active components in lumbricus was optimized; Using HPLC method, the fingerprint of the sample by the optimized technology was investigated, and the similarity of fingerprint was compared with the earthworm medicine, and the optimum freeze-drying process was obtained. Results: The best freeze-drying process conditions of active components in lumbricus were as follows: frozen temperature of -26.5 ℃, precool for 4 h, sublimation drying temperature of -20 ℃, sublimation time of 7 h, analytical drying temperature of 30 ℃, analytical drying time of 3.5 h, freeze-dried rate of 96.55%, and compared with medicinal materials of amino acid, the similarities of fingerprint were both greater than 0.9. Conclusion: Box-Behnken response surface method is used for the freeze-drying process conditions of active components in the lumbricus, the optimization is feasible, the effect of model is good, and this optimization process has the feasibility.

ABSTRACT

Objective To explore the accuracy of heart function assessment of the left single ventricle problems in children by three-dimensionalechocardiography (3 DE),the doppler myocardial performance index (MPI) and tissue doppler imaging(TDI).Methods Twenty-nine healthy children (the healthy control group) and 29 children with problems in the single left ventricles (the case study group) were enrolled in this study.End-diastolic volume (EDV),end-systolic volume(ESV) and ejection fraction(EF) were acquired by using IE 33 system and Q-lab software between 2 groups.MPI was calculated from doppler tracings on the ventricular inflow and outflow,and the velocity profile of atrioventricular annular was obtained by TDI.Max(dp/dt) of the left single ventricle was estimated by atrioventricular regurgitation using simplified Bernoulli equation.Results EDV,ESV and MPI in the case study group were higher than those of the healthy control group(t =2.821,5.287,2.085,all P <0.05),and EF of the case study group was lower than that of the healthy control group(t =-13.840,P < 0.05).Em,Am and Em/Am on the side of the left ventricular lateral wall of mitral annulus in the control group were higher than those of the case study group at the nonresidual cardiac cavity side(t =-3.059,2.219,-4.762,P < 0.05) ;Sm and Em/Am in the healthy control group at mitral annular lateral ventricular septal side were higher than those in the case study group at the residual cardiac cavity side(t =-2.200,-2.962,P <0.05).Q-Sm interval in healthy control group was lower than that in the case study group(t =2.345,P < 0.05).There was a negative correlation between Max (dp/dt) and MPI in the case study group (r =-0.57,P <0.05),there was a positive correlation between Max(dp/dt) and EF and Q-Sm interval in the case study group(r =-0.74,0.62,P < 0.05).Conclusions Impaired ventricular functions can be found in patients with left single ventricle problems,which can be assessed accurately by 3DE.

ABSTRACT

Objective To evaluate the diagnostic value of prenatal echocardiography in fetus with tetralogy of Fallot(TOF),and to analyze the reason of misdiagnosed in order to improve the echocardiographic diagnosis.Methods A retrospective echocardiographic review of 36 fetuses with TOF confirmed by prenatal magnetic resonance imaging (MRI) or postnatal echocardiography.Results Among 36 cases of TOF fetuses,31 cases (86.1 %) were diagnosed correctly by echocardiography,and in the remaining 5 cases,4 cases(11.1%) were misdiagnosed [2 cases misdiagnosed as ventricular septal defects (VSD),2 cases misdiagnosed as double outlet of right ventricles],and 1 case (2.8%) was missed diagnoses at 20 weeks gestation and confirmed as TOF at 36 weeks gestation based on the echocardiographic findings of malalignment type VSD,overriding aorta,pulmonary stenosis,and right ventricular hypertrophy.Malalignment type VSD,overriding aorta,and pulmonary stenosis were observed in 31 cases with TOF diagnosed by prenatal echocardiography,right ventricular hypertrophy was observed only in 6 cases.The most common associated anomalies were tricuspid regurgitation in 13 cases(36.1%),aortic insufficient in 6 cases(16.7%),absent pulmonary valve in 4 cases(11.1%),right-side aortic arch in 4 cases(11.1%),persistent left superior vena cava in 3 cases(8.3%),and complete atrioventricular septal defect in 1 case (2.8%).Conclusions Combining visualization of TOF in four-chamber view,outflow tract view,and three vessels view,TOF can be accurately diagnosed by prenatal echocardiography.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the association of occupational stress and related factors with type 2 diabetes mellitus (T2MD).</p><p><b>METHODS</b>In case-control study, a questionnaire survey was conducted in 201 T2MD patients and 201 controls, who were selected from the staff members of organizations, enterprises, and institutions in Ningxia Hui Autonomous Region, China according to inclusion and exclusion criteria, to acquire the information on general condition and occupational stress. These subjects also underwent physical examinations and blood biochemical analyses.</p><p><b>RESULTS</b>The T2MD group had significantly higher total occupational stress score, as well as the scores on such factors as workload, interpersonal relationship, and home/work balance than the control group (P < 0.01). After adjustment for age, gender, education level, smoking, and drinking, the odds ratios for T2MD were 2.538 and 3.075 in the people with moderate and severe stress, respectively, compared to those with mild stress. The risk factors for T2MD included drinking, family history of diabetes, waist circumference, triglyceride level, and total occupational stress score, while the protective factors included educational level and high-density lipoprotein level.</p><p><b>CONCLUSION</b>Occupational stress is associated with the incidence of T2MD; the higher the degree of stress, the greater the risk of T2MD. Relevant measures should be taken to reduce the occupational stress or improve the ability of workers to cope with the stress, thus decreasing the incidence of T2MD among occupational population.</p>

ABSTRACT

<p><b>BACKGROUND</b>Cyclooxygenase-2 (COX-2) is overexpressed during airway inflammation in chronic obstructive pulmonary disease (COPD) patients. Single nucleotide polymorphisms (SNPs) in the COX-2 promoter might contribute to differential COX-2 expression and subsequent interindividual variability in susceptibility to COPD. We investigated the association between COX-2 (-765G > C, -1195G > A) polymorphisms and COPD susceptibility in Japanese and Chinese patients.</p><p><b>METHODS</b>COX-2 genotypes were determined by polymerase chain reaction, restriction fragment length polymorphism in 230 COPD patients (103 Japanese and 127 Chinese) and 273 healthy controls (129 Japanese and 144 Chinese).</p><p><b>RESULTS</b>The frequency of -1195AA homozygote was significantly higher than the controls in Chinese COPD (adjusted OR = 2.43, 95%CI 1.14 - 4.19), Japanese COPD (adjusted OR = 2.25, 95%CI 1.06 - 4.76) and combined COPD groups (adjusted OR = 2.26, 95%CI 1.34 - 3.99). There was no difference in COX-2 -765G > C polymorphism between COPD and control groups in either Japanese or Chinese, while more Chinese individuals carried the -765C allele than Japanese in both groups (15.3% vs. 2.9% in COPD, 18.8% vs. 5.5% in control). Chinese individuals with the haplotype -765G:-1195A were at higher risk for COPD (adjusted OR = 1.93, 95%CI 1.05 - 3.55).</p><p><b>CONCLUSIONS</b>The COX-2 -1195AA genotype is associated with increased risk for COPD in both Japanese and Chinese individuals. Although COX-2 -765G > C polymorphism was not associated with COPD in either ethnic group, the -765C allele frequency was higher in Chinese than Japanese and haplotype -765G-1195A may confer susceptibility to COPD in Chinese.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the recent clinical safety and efficacy of photoselective vaporization of prostate (120 W) for the treatment of large gland benign prostatic hyperplasia.</p><p><b>METHODS</b>The clinical data of 112 cases who accepted photoselective vaporization of prostate (120 W) for the treatment of severe benign prostatic hyperplasia (the weight of prostate > 75 g) from July 2010 to January 2012 was statistical analyzed. Relief symptoms and complications were observed around surgery, and the recent clinical efficacy was analyzed.</p><p><b>RESULTS</b>All the operations were smooth. There were not transurethral resection syndrome. No cases need transfusion intraoperative and postoperative. The operation average time was (52.6 ± 12.1) minutes, and the average amount of bleeding was (27.4 ± 18.5) ml. The postoperative bladder irrigating time was (19.4 ± 7.3) hours, the mean postoperative indwelling catheter time was (3.2 ± 0.6) days, and the mean postoperative hospital stay was (4.8 ± 1.3) days. Postoperative international prostate symptom score (t = 52.24 - 59.10), quality of life (t = 48.42 - 53.63), maximum flow rate (t = -31.01 - -24.23) and residual urine volume (t = 9.85 - 12.53) compared with preoperative are significantly improved (P < 0.01).</p><p><b>CONCLUSIONS</b>With the safe operation of photoselective vaporization of prostate (120 W) for the treatment of large gland benign prostatic hyperplasia, it is less bleeding, recent efficacy is significant, and it is especially appropriate to elderly patients at high risk of large gland.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore operative complications of photoselective vaporization of prostate (120 W) for treatment of benign prostatic hyperplasia (BPH).</p><p><b>METHODS</b>The clinical data of 186 cases who underwent photoselective vaporization of prostate (120 W) for the treatment of BPH from May 2010 to April 2012, was statistically analyzed.</p><p><b>RESULTS</b>The operative time ranged from 7 to 147 minutes, and the average time was (37.7 ± 21.5) minutes. No patient accepted intraoperative blood transfusion, and occurred transurethral resection syndrome or capsular perforation. The time of postoperative indwelling catheter ranged from 1 to 11 days, and average time was (4.3 ± 2.2) days. Surgical outcome was satisfactory. Early postoperative complications included bladder spasm (3 cases), transient dysuria (19 cases), urinary tractirritation (94 cases), secondary hemorrhage (26 cases), transient urge incontinence (19 cases), all cases were relieved after treatment. Long-term complications, including recurrence (1 case), bladder neck stenosis (2 cases) and urethral stricture (2 cases), who had required reoperation. Postoperative patients with international prostate symptom score (29.4 ± 3.4), maximum urinary flow rate ((6.0 ± 1.6) ml/s) and residual urine ((167 ± 150) ml) had improved (t = -76.0 - 61.4, P < 0.01).</p><p><b>CONCLUSIONS</b>With less invasive, less bleeding and rapid postoperative recovery, photoselective vaporization of prostate (120 W) is a safe and effective minimally invasive treatment techniques for BPH. But there is still some complications after surgery and proper handling is required.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the etiology of acute hepatitis hospitalized patients in Beijing Ditan Hospital from 2002 to 2011.</p><p><b>METHODS</b>We summed up the changes in the characteristics of the etiology of acute hepatitis of patients mentioned above, and preliminarily analyze the causes.</p><p><b>RESULTS</b>From 2002 to 2011, 6235 patients with acute hepatitis were admitted to Ditan Hospital, aged between 12 and 78 years old, Of which 4309 were male and 1926 female. Acute viral hepatitis accounted for 70.44%-85.07%, while CMV, EBV, drug-induced liver injury accounted less than 5%, and acute hepatitis D and acute hepatitis C less than 1.10%. From year to year, the incidence and constitution of acute hepatitis changed significantly. The proportion of patients with acute hepatitis in total hospitalized patients was from 20. 38% to 2.05%. In 10 years, the percentage of acute hepatitis A decreased most obviously, about 99.11%, while 45.07% decline in incidence of acute hepatitis B and 62. 28% of acute hepatitis E. The constituent ratio of acute hepatitis also changed significantly. The proportion of acute hepatitis A declined from 31.31% in 2002, to less than 1% in 2011. The proportion of acute hepatitis B increased from 26.47% in 2002 to 45.88% in 2011, an increase of about 2 folds in 10 years. The proportion of acute hepatitis E increased from 26.73% in 2002 to 32.05% in 2010, a rise of 1.20 times in 10 years.</p><p><b>CONCLUSIONS</b>The proportion of patients with acute hepatitis in total hospitalized patients decreased from 20. 38% in 2002 to 2. 05% in 2011 in Beijing Ditan Hospital. The constituent ratio of acute hepatitis changed, too.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of social support and personality traits on psychological characteristic of patients with chronic cervicodynia and lumbodynia and improve the level of diagnosis and treatment.</p><p><b>METHODS</b>From August 2009 to April 2010, 231 patients (obtained 217 effective responses) with chronic cervicodynia and lumbodynia were recruited. Among the patients, there were 123 males and 94 females, with an average age of (38.00 +/- 5.67) years (ranged from 15 to 66 years). Social Support Rating Scale (SSRS), Cattell Sixteen Personality Factor Questionnaire (16PF) and Symptom Checklist 90 (SCL-90) were used to test social support and psychological characteristic and compared the difference of psychological, personality traits and norm, then analyzed the effect of social support and personality traits on psychological characteristic.</p><p><b>RESULTS</b>Two hundred and seventeen (93.9%) patients completed the questionnaire. Compared with normal 16PF scores, there were significant differences in factor scores of intelligence, stability, excitability, perseverance,social boldness, vigilance, sophistication, experimental, independence and tonicity (P < 0.01). And for SCL-90, the score of somatization, depression, anxiety, rivalrounsness, horror, bigotry, total score and mean score were higher than norm (P < 0.01). For SSRS, subjective support points and total points had positive effects; While intelligence, stability, perseverance, self-discipline had positive effects, vigilance and anxiety had negative effects.</p><p><b>CONCLUSION</b>There were significant differences in personality traits and psychological characteristic between patients with chronic cervicodynia and lumbodynia and norms. Improving social support level and optimizing personality traits can improve psychological profile of these patients.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the relation of serum HBsAg, HBeAg contents and HBV-DNA load changes in HBeAg-positive chronic hepatitis B during IFN-alpha treatment.</p><p><b>METHODS</b>After enrolled, the patients were treated with 3MU-5MU IFN subcutaneous injection every two days, and their serum was collected before treatment and every 3 months during the treatment course. The serum HBV-DNA load was determined by real-time fluorescence quantitative PCR method kit (lower detection limit 500 copies/ml, Piji company, Shenzhen city, China,) according to production instruction, and HBeAg and HBsAg contents were detected by ARCHITECH I 2000.chemiluminescent kit. The relation of serum HBV-DNA, HBeAg and HBsAg content was analyzed by SPSS statistic software.</p><p><b>RESULTS</b>There were 228 patients enrolled into this group, male 162 cases, female 66 cases, aged 14-60 years, average 30.94 years old. After IFN treatment the HBV-DNA, HBeAg and HBsAg levels were all gradually decreased. But there was no relation of HBsAg content to HBV DNA and HBeAg content before and during treatment course(P > 0.05). However the serum HBeAg content was related to HBV-DNA content significantly (P < 0.05) and their changes was correspondence.</p><p><b>CONCLUSION</b>Before and during treatment of interferon, HBeAg and HBV-DNA content changes are closely related, while there is no significant correlation between HBsAg and HBeAg and HBV-DNA content. During interferon therapy, HBsAg, HBeAg, HBV-DNA contents should be detected together.</p>

<p><b>OBJECTIVE</b>To evaluate whether the rapid viral response (RVR) to combinational therapy with interferon and rabavirin can be used to predict the sustained viral response (SVR) in chronic hepatitis C patients.</p><p><b>METHODS</b>According to their clinical characteristics, all patients in this study were given pegylated or conventional interferon injection and different dose of ribavirin according to their weight. Patients were injected Pegasys (pegierferon alpha-2a) 180 microg or 135 microg once a week, or pegyintron 50-80 microg once a week, or conventional interferon 3-5 MU every two days, in combination with a dose of 600-1500 mg/d ribavirin. The serum HCV RNA load was determined at 0, 4, 12 week, and then every 12 weeks. After the viral response obtained, the patients were treated for another 24-72 weeks and followed up 24 weeks. The main parameter to evaluate the efficacy was SVR rate. The influence factors associated with rapid viral response were investigated.</p><p><b>RESULTS</b>RVR was obtained at week 4 in 84.2% of the 120 patients. The HCV RNA baseline of RVR group was (5.883+/-1.246) lg copies/ml, which was significantly lower than that of the group without RVR [(6.502+/-0.693) lg copies/ml, t=2.15, P=0.034]. 97 patients with RVR who finished treatment and follow-up, 90.7% of these patients obtained SVR, but the SVR rate in patients (82.4%) without RVR was lower than that in patients with RVR (x2=0.371, P=0.543). In this study, RVR rate was not associated with HCV genotype and the dose of interferon used. In the naive patients, the RVR to pegylated interferon was 87.8%, which was significantly higher than that in retreat patients (x2=4.651, P=0.031).</p><p><b>CONCLUSION</b>High RVR rate could be obtained in chronic hepatitis C patients treated combined with interferon and ribavirin. RVR rate is associated with the HCV RNA baseline load in both naive and retreat patients but not correlated to HCV genotype. RVR could predict the SVR.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effect of arsenic trioxide (As2O3) on the growth inhibition of imatinib (IM)-resistant bcr-abl mutant cell lines in vitro.</p><p><b>METHODS</b>Cell growth of one IM-sensitive cell line, 32Dp210 and 15 IM-resistant cell lines including T315I and other 14 bcr-abl mutants were detected by MTT assay after treatment with IM and As2O3. The cell lines with 5 frequently observed mutants in CML patients were analyzed for apoptosis by flow cytometry with Annexin V and PI staining as well as the expression of bcr-abl fusion protein, phosphorylated CRKL protein and apoptosis-related proteins by Western blot.</p><p><b>RESULTS</b>The fifty percent inhibition concentration (IC50) values of As2O3 for 15 IM-resistant cell lines were 2.6-5.3 fold lower than that for IM-sensitive cell line. For the 5 bcr-abl mutants frequently happened in CML patients, As2O3 significantly inhibited the expression of bcr-abl fusion protein and phosphorylated CRKL and induced apoptosis in a dose-dependent manner as compared with that for 32Dp210. Coincidently, the cell apoptosis was induced through caspase-3, 8 and 9 pathways.</p><p><b>CONCLUSION</b>As2O3 remarkably inhibits cell growth and induces apoptosis of IM-resistant bcr-abl mutant cell lines in vitro, suggesting that it might be a potential therapeutic agent for IM-resistant CML patients.</p>

ABSTRACT

This study was aimed to compare HHGV678 with imatinib (IM) in growth inhibition of Bcr-Abl wild type and IM-resistant cell lines, investigate the possibility of replacing IM with HHGV678 in treatment of chronic myeloid leukemia (CML) and IM-resistant CML patients. Viability of two Bcr-Abl wild type cell lines (K562 and 32Dp210) and 16 IM-resistant cell lines (K562R and 15 Bcr-Abl point mutant cell lines) treated with HHGV678 and IM was analyzed by MTT. The apoptosis of those cells was identified by flow cytometry with Annexin V staining and DNA ladder analysis. Western blot was applied for detecting the expression of Bcr-Abl and phosphotyrosine protein levels. The results indicated that HHGV678 significantly inhibited the growth of two Bcr-Abl wild types and IM-resistant cell lines in dose-dependent manner except cell line of T315I point mutant. IC(50) results showed that the growth inhibition of HHGV678 was 15.5 and 28-fold higher than that of IM in K562, 32Dp210 and 1.4 to 124.3-fold higher than that of IM in 15 IM-resistant cell lines respectively. Compared with IM, HHGV678 more significantly inhibited phosphotyrosine kinase protein of the cells mentioned above at different concentrations. With most importance, HHGV678 of 10.0 micromol/L induced cell apoptosis of 40.06% and 33.32% in K562R and 32Dp210(T315I) cell lines, which were much higher than that of IM (19.77% and 10.68%). It is concluded that HHGV678 is more effective than IM in the growth inhibition of Bcr-Abl wild type cell lines and IM-resistant cell lines, especially in strongest IM-resistant cell lines. Further studies are needed to show whether HHGV678 may be a novel targeting drug in treatment of CML and IM-resistant CML patients.

ABSTRACT

<p><b>OBJECTIVE</b>To understand the action mechanisms of artesunate on inhibiting leukaemia cell line K562 on the molecular level.</p><p><b>METHOD</b>The gene chip was used to detect the expression panel of genes of leukaemia cell line K562 treated by dihydroartemisinin. K562 cells were treated with 1 x 10(-5), 4 x 10(-5), 16 x 10(-5), 64 x 10(-5), 256 x 10(-5) mol x L(-1) dihydroartemisinin for 24 h, and then studied the modality changes by invert microscope. The morphological changes of the nucleons were observed by Hoechst33342/PI staining. The cell cycle were examined by flow cytometry analysis (FCM). Total RNA samples were obtained by TRIzol and were reverse transcribed to the cDNA. The cDNA samples were hybridized to our gene chips. Hybridization signal were collected and analyzed following scanning by Gene Pix 4100A.</p><p><b>RESULT</b>The numbers of drift cells were increased and the density of cells was decreased under invert microscope after K562 cells were treated with dihydroartemisinin for 24 h. Morphological changes of cell apoptosis such as karyopyknosis and conglomeration were observed by Hoechst 33342/PI staining. Flow cytometric analysis showed that cells were arrested in G2 phase. There were 13 differentially expressed genes identified. Hybridization analysis showed up-regulation of chk1 and down-regulation of PCNA, cyclinB1, cyclinD1, cyclinE1, cdk4, cdk2, E2F1, DNA-PK, DNA-Topo I, mcl-1, jNK, VEGF in the dihydroartemisinin-treated K562 cells.</p><p><b>CONCLUSION</b>Dihydroartemisinin can Inhibit the leukaemia cell line K562 and exert its anti-cancer effect by altering the expression of these genes involved in cell cycle; dihydroartemisinin may act via apoptosis pathway.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the influence of hepatitis C virus (HCV) serotype on the interferon (IFN) treatment of patients with chronic hepatitis C.</p><p><b>METHODS</b>Ninety-eight patients with chronic hepatitis C were divided into two groups: patients in group 1 (n=53) were treated with Pegasys, 180 ug injected subcutaneously once a week for 24 weeks, and those in group 2 (n=45) were injected with Roferon-A 3 MU three times a week for 24 weeks and then patients in both groups were followed up for another 24 weeks. The virological response at the end of follow up was the primary endpoint for evaluating the effects of IFN treatment. The HCV RNA levels of the chronic hepatitis C patients were determined with COBAS AMPLICOR MONITOR Test, version 2.0, and the HCV serotypes were examined by the means of ELISA using Murex HCV Serotyping 1-6 Assay.</p><p><b>RESULTS</b>Of the 98 cases, HCV in 44 cases was serotype 1, in 23 was serotype 2, in 10 was serotype 3, in 1 was serotype 4, 1 was serotype 5 and in 2 was serotype 6; HCV serotypes in the remaining 17 patients could not determined. In Pegasys treatment group, the biochemical and virological response was not significantly different at the end of treatment between the patients with serotype 1 and non serotype 1 or serotype undetermined patients, but the sustained virological response rate of HCV serotype undetermined group (66.7 percent) was significantly higher than that of serotype 1 patients (27.3 percent) (p=0.035). In Roferon-A treatment group, the virological response rate at 24 weeks and sustained viral response rate at the end of follow-up was not significantly different between serotype 1 and non serotype 1 patients or serotype undetermined patients.</p><p><b>CONCLUSION</b>After the six months treatment course, the HCV serotype had some effects on the treatment response to Pegasys treatment for chronic hepatitic C.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the tested results of the segmental range of motion (ROM) of lumbar spine by charge couple device (CCD)-based system for 3-dimensional real-time positioning (CCD system), and to analyze its clinical significance.</p><p><b>METHODS</b>Seven patients with lumbar joint dysfunction and 8 healthy subjects were tested twice by the CCD-based system with an interval of 10 min.</p><p><b>RESULTS</b>The ROM of the patients was obviously lesser than that of the healthy subjects. The measuring data of segmental ROM of lumbar spine by CCD system is correlated significantly to the same data checked later on the same subjects in every direction of the movements. The differences between two checks are usually less than 1 degree.</p><p><b>CONCLUSION</b>Specially designed CCD based system for 3-dimensional real-time positioning could objectively reflect the segmental ROM of lumbar spine. The system would be of great clinical significance in the assessment of the biomechanical dysfunction of lumbar spine and the effect of the treatment applied.</p>
